Study of Oxidative Stress Induced by Okadoic Acid and Its Effect Brain Antioxidant System in Zebra Fish (Danio Rerio)

Abstract
Alzheimer's disease (AD) is a neurological illness associated with aging. To learn more about AD mechanisms and aid in medication discovery, transgenic and pharmacological AD models are researched in great detail. Nevertheless, they are time-consuming and somewhat expensive, which makes it more difficult to find promising anti-AD treatments. Here, we created a novel model of AD in adult zebrafish by co-treating them for 72 hours with okadoic acid (OKA) at varying concentrations. Following treatment we studied oxidative stress using Superoxide dismutase (SOD), Glutathione Peroxidase assay (GPX), Acetylcholinesterase (AChE) activity and DNA fragmentation. We found the brain weight of the treated groups was found to be reduced along with AChE activity. The activity of glutathione reductase (GR) and SOD activity got increased with OKA concentrations and also we did observe prominent DNA shearing with increasing concentrations of OKA. Based on the findings, we may infer that OKA adversely affects antioxidant biochemical pathways linked to the generation of reactive oxygen species in the zebra fish that were studied.
